Thyroid peroxidase (TPO) is a 100-kDa poorly glycosylated membrane-bound enzyme containing a heme prosthetic group. The enzyme is expressed on the apical membrane of thyrocytes, facing the colloid, where it is responsible for catalyzing iodine oxidation, iodination of tyrosine residues and coupling of iodotyrosines to generate thyroid hormones . TPO is identical with the previously defined thyroid microsomal antigen , and measurement of TPO-Ab has replaced measurements of antibodies against the microsomal antigen (Mic-Ab).
