Thyroid peroxidase (TPO) is a glycosylated heme-protein located in the apical membrane of the thyrocyte, where it catalyzes iodide oxidation and further incorporation in the thyroglobulin molecule. TPO shares sequence homology with lactoperoxidase, myeloperoxidase and eosinophil peroxidase. Two isoforms, hTPO-1 and hTPO-2 have been isolated, the latter being truncated and devoid of enzymatic activity. TPO is also a major thyroid antigen, to which the immune response is directed whenever tolerance to thyroid is disrupted. TPO variants have been associated with autoimmune hypothyroidism
