BNP was originally isolated from the porcine brain, but its main site of synthesis, storage, and release is, like ANF, in the atria of the heart. BNP-binding sites are found in the subfornical organ, supraoptic nucleus, and paraventricular nucleus. BNP was shown to compete with the majority of the high-affinity binding sites for ANF in the rat’s brain. Immunohistochemically, BNP is localized in the hypothalamus of the rat and the cynomolgus monkey. These findings suggest that BNP in concert with ANF is involved in CNS mechanisms for modulating cardiovascular homeostasis.
